Comparison and Analysis of Mitochondrial DNA in cytoplasmic Male Sterile brassica napus



Abstract
Mitochondrial DNA (mtDNA) was isolated and compared in normal and cytoplasmic male sterile (CMS) lines of Brassica napus. A plasmid-like mtDNA was detected at about 11.3 kb only in the CMS line. In the Southern hybridization analysis of Hind III-digested mtDNA, by using the [¥á^(32)-P]-labelled ATP¥á gene and CoxI gene as probes, only one band for each line was detected without regard to the cytoplasmic origin, one band at about 8.0 kb in CMS lines and a band at about 5.3 kb in normal line, respectively.
